KUP101 takes a different approach to many established cancer immunotherapies. The active substance simultaneously activates Toll-like receptors (TLR) 4 and 7, thereby mobilising the innate immune system against tumours. In addition, Kupando targets what is known as ‘trained immunity’. This involves modifying cells of the innate immune system so that they can respond more strongly to further stimuli. The study is initially investigating safety and tolerability, as well as pharmacokinetics and pharmacodynamics. It includes patients with advanced solid tumours, with a focus on various types of skin cancer. A total of 21 participants are planned. Following the first treatment, a safety committee has approved the enrolment of further patients. KUP101 is also of interest with regard to combination therapies. In preclinical studies, Kupando observed additional effects when used in combination with checkpoint inhibitors. Clinical development must now demonstrate whether these results can be extrapolated to patients. For Kupando, the start of the trial is also a key value driver. The company raised a further 10 million euros in March, thereby increasing its Series A funding to a total of 23 million euros.
